If you have an LTI system with an input signal $x(t)$ and an output signal $y(t)$ the transfer function can be found as
$$H(\omega) = \frac{\mathcal{F}\{y(t)\}}{\mathcal{F}\{x(t)\}}$$
where $\mathcal{F}\{\}$ denotes the Fourier Transform.
